This article explains the scientific considerations that informed the latest evolution of the AgeMate formula.
NMN Remains the Foundation
Although the formulation has evolved, one thing has not changed. NMN remains the foundation of the AgeMate formula at 500 mg per serve. Human clinical studies have investigated NMN across a range of doses, generally between approximately 100 mg and 1,200 mg per day. These studies have reported that supplementation is generally well tolerated and can increase NAD+ levels (R,R).
What’s New in the Formula
Rather than simply increasing the number of ingredients, the updated formulation reflects an ongoing review of emerging research, ingredient functionality, manufacturing considerations and regulatory requirements.
Added: Dimethylglycine (DMG) — 50 mg
One of the new additions is dimethylglycine (DMG). DMG participates in normal one-carbon metabolism, contributing methyl groups used in methionine metabolism and homocysteine recycling.
As understanding of nicotinamide metabolism has evolved, maintaining adequate methylation support has become an increasingly discussed consideration within longevity research. DMG was included to complement the formulation’s broader methylation nutrient network alongside folate and vitamin B12 (R).
Added: Biotin and Zinc
Biotin and zinc were introduced as part of the revised formulation following the removal of oral hyaluronic acid. Together with vitamin C, these nutrients support recognised physiological functions associated with connective tissue, skin health, immune function and DNA synthesis (R, R, R).
Updated to Better Reflect Current Research
L-Theanine
Published human studies investigating L-theanine for relaxation, attention and stress management have commonly used doses of approximately 200 mg. Increasing the amount to 200 mg brings the AgeMate formulation more closely into line with the dosage investigated in much of the published clinical literature (R).
Glycine
Glycine is an amino acid involved in collagen synthesis, glutathione production and normal metabolic processes. Emerging research has also investigated glycine in relation to healthy ageing, metabolic health and mitochondrial function, although research in these areas continues to develop (R, R). Increasing glycine to 1.1 g reflects the growing scientific interest in this ingredient and strengthens its contribution to the overall nutrient profile.
Why We Removed Certain Ingredients
Responsible formulation also involves reviewing whether every ingredient continues to align with the overall formulation strategy.
Spermidine
Spermidine has attracted considerable research interest within longevity science. However, human evidence relating to supplemental spermidine continues to evolve. It is also not currently permitted for use in Australian TGA-listed medicines. Following this review, we chose to focus on ingredients that better aligned with both the current regulatory framework and the broader formulation strategy.
Pterostilbene
Pterostilbene was removed because it is not currently permitted in Australian TGA-listed medicines. As a fat-soluble compound, it was also less compatible with the water-based powdered format used in the AgeMate formulation.
Hyaluronic Acid
Oral hyaluronic acid was removed as part of formulation optimisation. Although it has been studied for skin hydration and joint health, the evidence remains more variable than for nutrients with established physiological roles. The revised formulation focuses on ingredients such as vitamin C, zinc and biotin, which have well-established functions in supporting normal connective tissue and skin health.
Vitamin B6
Vitamin B6 was removed as part of formulation optimisation. While it remains an important essential nutrient, it was not considered to provide a distinct functional contribution within this formulation relative to the remaining ingredients. This approach ensures each ingredient has a clear scientific rationale and supports the intended purpose of the finished product (R).
Natural Colours, Better Ingredients
The updated formula replaces butterfly pea with naturally derived blue spirulina and beetroot. We have also selected thaumatin, a naturally derived sweet protein, as the sweetener. These decisions reflect our preference for ingredient choices that support the overall formulation while maintaining quality and consistency.
